Regeneration strategy of five types of liquid chromatography
The regeneration of chromatographic column is usually realized by recoil. The chromatographic column is reversed connected and disconnected from the detector. The following solvent is used to recoil the chromatographic column. The column pressure is controlled in the range of 1500-4500psi. The standard sample can be used to evaluate the performance of the column after the column regeneration.Silica gel column:
50ml n-hexane
50 ml dichloromethane
50ml isopropanol
50ml methanol
25ml dichloromethane
25ml mobile phase
Silicon based reverse column (the commonly used C18 column belongs to this type)
There are two kinds of chromatographic columns, one is to remove the water-soluble sample by recoil, and the other is to remove the sample insoluble in water by recoil.
The sample flow in water is as follows:
Rinse with deionized water at 60 ℃
50ml methanol
50ml acetonitrile
25ml methanol
25ml mobile phase
The process flow of water-insoluble sample is as follows:
50ml isopropanol
50 ml dichloromethane
50ml n-hexane
25ml isopropanol
25ml mobile phase
Polar column
50ml chloroform
50ml methanol
50ml acetonitrile
25ml dichloromethane
25ml methanol
25ml mobile phase
Silica based ion exchange chromatographic column
Wash with 50 ml deionized water at 40 ~ 60 ℃
50ml methanol
50ml acetonitrile
25ml dichloromethane
25ml methanol
25ml mobile phase
Cyanopropyl bonded chromatographic column
Remove the protein
The chromatographic column was washed with acetonitrile and water solution containing 0.1% trifluoroacetic acid at a ratio of 1:1;
Remove the TCA
1) Distilled water 10 times the volume of column, adjust pH to 2.5 with phosphoric acid (main function is to remove salt)
2) 10 times the volume of column methanol (the main function is to remove water)
3) 10 times column volume of methanol and dichloromethane 1:1 solvent (cleaning solvent)
4) Methanol with 10 times column volume
Note:
Today, all volume values are calculated with a chromatographic column with an inner diameter of 25cm * 4.6mm. For chromatographic columns not of this specification, the corresponding volume needs to be converted. Different lengths are directly converted by proportion, and different inner diameters are converted by the square ratio of diameter.
