Packing description
 
   The matrix of target-Gly filler is polystyrene divinylbenzene (PS/DVB) or Polymethylmethacrylate(PMMA) spherical particles. Using special chemical bonding technology, ion exchange functional groups are uniformly and finely bonded on the surface of polymer hydrophilic layer. 
 
Packing stability
 
  Because the surface modification of PS/DVB and PMMA is completely based on chemical bonding, Target-Gly ion exchange filler has excellent stability. The stationary phase can be compatible with most buffers such as ammonium acetate, phosphate, Tris, etc. At pH 6.0, Target-Gly column was operated with 20 mM phosphate buffer solution as the mobile phase, and the column efficiency hardly changed.
 
Mobile phase compatibility
 
  Compatible with aqueous solution, mixture of water with acetonitrile, acetone or methanol, etc. Typical buffers include phosphate, Tris and acetate. 
 
Flow rate
 
  The typical operating flow rate of chromatographic column with inner diameter of 3.0-4.6mm is 0.40-1.0ml/min.
 
Installation and operation of  the  column
 
   Both ends of the chromatographic column are always sealed with plugs during transportation or when not in use. When connecting the chromatographic column to the chromatographic instrument system, first remove the plugs at both ends. Please note that the flow direction of the mobile phase is consistent with the direction marked on the column. Unless for special consideration, for example, when the chromatographic column needs to be connected reversely for flushing in order to remove the dirt blocked at the inlet of the chromatographic column, it is recommended that the user must follow the direction marked on the column when connecting the chromatographic column. Since the connection of chromatographic column is a part of the whole chromatographic operation process, if the sealing ferrule is too tight or improperly installed, or the sealing ferrule does not match the chromatographic column port, it may lead to the leakage of solution. Please connect the chromatographic column with the sealing ferrule according to the following steps to connect the chromatographic column to the HPLC system:
  
(1) For the pipeline used for the first time, please put the pipeline joint and sealing card on the pipeline with an outer diameter of 1 / 16 ". The wide end of the sealing ferrule shall face the pipeline joint.
 
(2) Tightly insert the pipeline into the interface of the chromatographic column, slide the sealing ferrule and pipeline connector forward, connect the thread of the pipeline connector with the thread of the chromatographic column port, and then tighten the pipeline connector. If the pipeline is made of polymer material, please go to step (4); If it is a metal pipeline, continue (3).
 
(3) After firmly pressing the pipeline into the column end interface, further tighten the tightened nut with a 1 / 4 "wrench.
 
(4) The other end of the chromatographic column is operated by the above method.
 
Sample and mobile phase
 
In order to avoid the blockage of chromatographic column, all samples and solvents, including buffer solution, must be filtered with 0.45um or 0.2um filter membrane before use. It is recommended to use pre- column filter (0.5m pore size) or protective column to protect the chromatographic column. Target-Gly chromatographic column can use aqueous solution or mixture of organic solvent (such as methanol, acetonitrile) and water as mobile phase. A typical mobile phase contains sodium or potassium salts of phosphoric acid, acetic acid or Tris. The mobile phase needs to be degassed before use. A simple degassing method is to ultrasonic the mobile phase under the vacuum formed by the water pump for 5 minutes.
 
Use of chromatographic column
 
     1) The transport solvent of the new Target-Gly column is 20 mM phosphate buffer (pH 6.0). During storage and transportation, the filler may dry up. When used for the first time, it is recommended to rinse the column with a transport solvent 10-20 times the column volume to activate the column. Then, the chromatographic column can be washed with the mobile phase selected by the user for balance. The flow rate gradually rises from 0.1ml/min to the required operating conditions until the baseline is stable. If the column pressure and baseline fluctuate greatly, it may be that bubbles enter the chromatographic column. At this time, the chromatographic column can be washed at a higher flow rate for 3-10 minutes. For example, the flow rate of 4.0 * 33mm chromatographic column can be 1.5-2.0ml/min. If the type or pH of the mobile phase is quite different from the storage solvent in the chromatographic column, it is recommended to wash the chromatographic column with a new mobile phase for 10 times the column volume.
2) In order to obtain the best separation effect and prolong the service life of the column, please try to use the mobile phase with pH in the range of 2-10.
3) The normal operating pressure should be less than 3500psi
4) The maximum operating temperature is 80℃. In order to prolong the service life of the column, please operate within 60℃ as far as possible.
5) When not in use for a long time, Target-Gly column should be stored in 20 mM phosphate buffer (pH 6.0) containing 0.1% NaN3. The chromatographic column needs to be flushed with storage buffer solution at least 20 times the column volume. Then plug both ends of the chromatographic column with two detachable plugs attached to the chromatographic column to prevent the column bed from drying up.
